A fluorescence microscope observation on self-pollination and cross-pollination of pollen tubes in Camellia oleifera‘ Min 48' and ‘Min 43'

Abstract

Using‘Min 48',‘Min 43'as materials,control pollination with fluorescence microscopy observation was combined to investigate pollen tubes growth differences between self-pollination and cross-pollination after pollination. The results showed that pollens by self-pollination and cross-pollination germinated normally,while pollen tubes growth rates were different in different period. The cross-pollination growth rate showed an‘up-down-up'tendency,appearing 2 extreme value( up to 67 μm·h- 1) in the period of 6- 7 h and 24- 30 h separately. Then the pollen tubes penetrated style base in 76 h after pollination. Comparing with cross-pollination,self-pollination presented a wave growth tendency: lag phase occurred in 6 h after pollination,then recovered growth in 9 h after pollination. A step rise happened in 24 h after pollination. Self-pollination pollen tubes kept a rapid growth tendency until they reached the peak value( 32. 22 μm·h- 1) in the period of 30- 48 h. In the final,the pollen tubes stopped growing and couldn't penetrate style base for callose blockage in pollen tube tip in 127 h after pollination.
