Abstract
OBJECTIVE To observe the growth inhibitory effect of Oxaliplatin( L-OHP) and Triptolide( TPL)on proliferation in human colon carcinoma cell lines SW480 in vitro and investigate the mechanism. METHODS The growth inhibitory effect was evaluated by MTT assay in single agent or combination treatment. Distribution of cell cycle and apoptosis was analyzed by using flow cytometry. RESULTS Combination treatment of TPL and L-OHP has synergistic effect on killing SW480 cells. The analysis of cell cycle indicated that TPL and L-OHP blocked cells at S phases and cells of G2/M phase reduced. CONCLUSION Combination treatment of TPL and L-OHP could inhibit the proliferation of SW480 cells obviously according to the experiment,which mechanism probably is a synergistic interaction in the cell cycle.
