Abstract
Aim: To investigate the effect of topotecan on the expression of Survivin in ovarian cancer cell. Methods:SKOV3 cells were cultured with different concentration of topotecan(0.244 μg/L,3.906 μg/L,62.5 μg/L,1 mg/L,16 mg/L,128 mg/L),then the Survivin protein and mRNA expression of SKOV3 were investigated by immunohistochemistry and RT PCR, respectively. Results:Under the effect of topotecan, expression of Survivin mRNA and protein significantly decreased, which showed a dose effect response. Compared with control group, the difference was significant ( P 0.05). As time and concentration of TPT increased, the expression of Survivin mRNA and protein were inhibited distinctly. Conclusion: TPT may inhibit expression of Survivin mRNA and protein in SKOV3 cells.
