Extraction and purification of polysaccharides from Pleurotus citrinipileatus sing

Abstract

Using the methods of single factor test and orthogonal design test of L9(33) ,the effects of various factors such as the extraction solvent,the ratio of sample to water,the extraction time,extraction temperature and extraction times on the extraction efficiency of polysaccharides in Pleurotus citrini-pileatussing were investigated and the optimal conditions were concluded. The polysaccharides were purified by removing protein,ethanol precipitation and AB-8 macroporous resin. The results showed that the main factors from the primary to the secondary affecting the extraction efficiency of polysaccharides were as follows:extraction time,extraction temperature and ratio of sample to water. The optimal conditions of extraction were established as follows:ratio of sample to water 1∶25,extraction temperature 90 ℃,extraction time 3 h. Under the optimized conditions,the extraction efficiency of polysaccharides in Pleurotus citrinipileatussing is 30.48%. The content of polysaccharide after purifying was 91%,the molecular weight was about 104356.
