Abstract
The immobilized lipase was prepared by using the glutaraldehyde to cross-link lipase and the CA modified by amino-silane coupling agent.The immobilization conditions were optimized by studied the effect of the glutaraldehyde concentration,activation time,enzyme quantity and crosslinking time.The results show that the optimal conditions are as follows;5% glutaraldehyde,activation 8 h,4 mL concentration of 1.64 mg/mL of lipase solution,at 4℃crosslink 10 h.Then the properties of the free lipase and the immobilized lipase were tested.The results show the storage stability,the adaptability for temperature and pH of the immobilized lipase excel to the free lipase.The immobilized lipase can use in more times,that can overcome the limitation of the free lipase and establish the foundation for the farther application.
