Abstract

ObjectiveTo investigate the expression of Cyclin E,retinoblastoma(Rb) gene protein and S-phase kinase associated protein 2(Skp2) in cervical carcinoma and cervical intraepithelial neoplasia(CIN),and to explore the significance of their expression in the carcinogenesis of cervical carcinoma.MethodsCyclin E,Rb and Skp2 were investigated with immunohistochemical staining(SP) in 77 cases of CIN(CIN Ⅰ 20 cases,CIN Ⅱ 20 cases,CIN Ⅲ 37 cases) and 40 cases of cervical invasive squamous cell carcinoma,and 20 cases of normal cervical epithelia were included as controls.ResultsThe positive rates of Cyclin E increased from normal cervical epithelia,CIN Ⅰ,CIN Ⅱ,CIN Ⅲ to cervical squamous cell carcinoma groups,(0.0 %),(30.0 %),(75.0 %),(65.0 %),(60.0 %),respectively and the positive rates of Skp2 in the above groups were(45.0 %),(80.0 %),(70.0 %),(94.6 %),and(92.5 %),respectively.The positive rates of Rb declined from normal cervical epithelia,CIN Ⅰ,CIN Ⅱ,CIN Ⅲ to cervical squamous cell carcinoma,(100.0 %),(55.0 %),(20.0 %),(24.3 %) and(23.5 %),respectively.The expression of Cyclin E in cervical squamous cell carcinoma and CIN was significantly higher than that of normal cervical epithelia.The expression of Skp2 in cervical squamous cell carcinoma and(CIN Ⅲ) was significantly higher than that of normal cervical epithelia.ConclusionThe positive rates of Cyclin E and Skp2 become higher and higher from normal cervical epithelia,CINⅠ,(CINⅡ),(CINⅢ) to cervical invasive squamous cell carcinoma,which suggests that their over-expression might play important role in the carcinogenesis of cervical carcinoma.The expression of Rb protein,however,becomes weaker from normal cervical epithelia,CINⅠ,CIN Ⅱ,CIN Ⅲ to cervical invasive carcinoma.The functional deactivation of Rb might take an important role in the carcinogenesis of cervical carcinoma.In gynecological pathology,the feature of Cyclin E and Skp2 expression could be helpful for the diagnosis of CIN and cervical carcinoma.
