Abstract
A method for γ-Amino Butyric Acid(GABA) determination in mature vinegar with ortho-phthaladehyde(OPA) automatic pre-column derivation and fluorescence detection by high performance-liquid chromatography was established.Firstly mature vinegar was pretreated with trichloroacetic acid of 10%,derived OPA and 9-fluorenylmethyl chloroformate(FMOC-CL),and then separated and assayed with reversed phase C18 column,finally eluted at 40℃ and determinated by HPLC at EX:250nm,EM:410nm.The result showed that GABA in mature vinegar was separated well with this method,and in the range of 0.004~0.30mg/mL of calibration curve,GABA kept linear with regression equation of Y=26016X-38.72(R2=0.9994).The relative error was 96.22%,which indicated that method was a sensitive,reproduced and specific assay for the determination of GABA in mature vinegar.
