The clinical research of cytokine-induced killer cells on the clinical immunotherapy for breast tumor

Abstract

Objective To investigate the cytokine-induced killer cells(CIK cells) cytotoxic activity in vitro,and to observe the short-term clinical efficacy,immunological activity and side effects of CIK cells in treating breast tumor.Methods A total of 35 patients with breast malignancies were recorded with CIK cell therapy from July 2007 to March 2010.Peripheral blood mononuclear cells(PBMC) from patients with standard treatment protocols were induced to CIK cell in vitro,then phenotype of CIK cells were observed and vitro cytotoxicity was measured by MTT method.We evaluated their clinical anti-tumour efficacy,immunological activity and side effects.Results Cytotoxic activity of CIK cells in vitro was 77.06±11.62 when the ratio of effector cell to target cell was 16:1.In the short-term clinical efficacy evaluation,partial remission ratio was 85.71%,and the disease control rate was 88.57%.No tumor progressing period was 2 to 31 months and the median was 15 months.The cells surface markers CD3+,CD4+ and CD8+ of patient's PBMC had increased(P0.001) compared with that before transfusion.It meant that patient's specific immune response could be induced by CIK cells.One case had fever at 37.5℃ during the infusion of CIK cells.Conclusion In breast tumor immunotherapy,CIK cells can induce specific immune response.It is a new anti-tumor effector cell,which has good clinical efficacy.
