Abstract

Objective To study the apoptosis-induced effect of the recombinant solubility TRAIL(rsTRAIL) on cell lines MG-63,and investigate the combination anti-tumor effect of rsTRAIL and cisplatin(CDDP). Methods The morphology of cultured cell treated with or without rsTRAIL was observed and compared microscopically.To explore the methods to increase the sensitivity to TRAIL,cancer cell lines MG-63 were treated with lower dosage of rsTRAIL and sub-toxicity level CDDP.The combination anti-tumor effect determined by MTT assay and AO/EB double fluorescence staining were compared with that in control groups treated with single TRAIL or CDDP. Results Cells killed by rsTRAIL show typical apoptosis morphology changes.The recombinant TRAIL had significant anti-proliferative effects on MG-63 cells.The anti-proliferative rate was 10.1% to 84.6%,and the apoptosis rate was 19% to 69.7%.The classic dose-effect relationship was founded.The sub-toxicity level CDDP can significantly increase the sensitivity of MG-63 to the recombinant TRAIL.Treated with CDDP and TRAIL,the apoptosis rate of MG-63 reached 81%. Conclusion The recombinant TRAIL can kill MG-63 cells by inducing cells apoptosis.CDDP can synergitically enhance the effect of the recombinant TRAIL on inducing apoptosis in A549 and MG-63 cells.
