Culture and chondrogenesis of mesenchymal stem cells derived from human bone marrow in vitro

Abstract

Objective: To research the phenomenon and rule of multiplication and chondrogenesis of mesenchymal stem cells(MSCs) derived from human bone marrow in vitro.Methods: Healthy adult′s MSCs were isolated from Ilium and purified by density gradient.To observe the conformation,growth and multiplication of MSCs cultured in vitro.We chose the passage 3 MSCs and induced them into chondrogenic differentiation with Dexamethasone,Vitamin C and TGF-β1 in different dosage formed chondracytes induction.After 3 weeks,we measured the sulfate glycosaminoglycan with toluidine blue and alcian blue,type Ⅱ collagen with immunofluorescence and glycosaminoglycan with alcian blue colorimetry. Results: MSCs can be cultured well in vitro.MSCs were induced into chondrocytes by TGF-β1 and TGF-β1 10 μg/L group had the most evident expression. Conclusion: MSCs have strong ability of multiplication and could be induced into chondrocytes under certain circumstances.TGF-β1 is the major factor of chondrogenesis while 10 μg/L is the best inducing dosage.
