Abstract
Objective To elucidate the expression of fragile histidine triad(FHIT) protein by western blotting array.Methods 30 human colorectal carcinoma specimens and adjacent normal colorectal mucosal tissues were determined by mole pathologic method.The expression of FHIT protein was detected by Western blotting array.Results 56.7% of the carcinomas showed a marked loss or absence of FHIT expression compared with their matched normal mucosas Carcinomas with reduced expression of FHIT correlated with their histological grade,Dukes'stage(P0.05).The distribution of decreased expression of FHIT was 3/11 in grade Ⅰ,carcinoma,8/12 in grade Ⅱ,6/7 in grade Ⅲ,respectively.The correlation between decreased expression of FHIT and Dukes' staging was 6/17 in stage A and B,11/13 in stage C and D.The difference between stage A,B with no lymph nodes metastases and the stage C、D with lymph nodes metastases was of significance(P0.05).Conclusion The western blotting array is an ideal method in detecting FHIT protein for it's differently and sensitive.The reduced expression of FHIT may be associated with decreasing differentiation,metastasis in colorectal carcinomas.It is suggested that decreased FHIT expression plays an important role in the development and progression of the tumor.
