Abstract

Two different sources of proteinase were combined, and used to hydrolyze porcine hemoglobin. The hydrolysis rate was regarded as the index of examination. The effects of hydrolysis conditions on the hydrolysis rate were considered,which were the substrate concentration, proportion of enzymes, pH, hydrolysis temperature, enzyme concentration and hydrolysis times. According to the orthogonal experimental design, the optimum hydrolysis condition of porcine hemoglobin by combinative enzyme treatment are as follows: the substrate concentration 10%, proportion of enzymes 3∶7, pH 7.5, hydrolysis temperature 50℃, enzyme con-centration 8000U/g, hydrolysis times 10h. On this condition, the hydrolysis rate was 19.1%.
