Effect of Serum Containing Tougu Xiaotong Capsule on Activity of Chondrocyte

Objective To observe the effect of serum containing Tougu Xiaotong capsule on the activity of chondrocyte.Methods Cartilage was isolated from the knee joint of SD rats,and used to establish a cultivation system of chondrocytes in vitro.The 3rd generation of chondrocytes were intervened after synchronization to determine the effective intervention time and intervention concentration.The chondrocytes were intervened by serum containing Tougu Xiaotong capsule under effective intervention concentration,and the proliferation of chondrocytes was observed at effective time points by MTT assay.Results After intervention of 72 h,the OD value of chondrocyte was significantly higher than that after intervention of 24 h,48 h and 96 h respectively(P0.05).The OD value of chondrocyte in 10% drug-containing serum group was significantly higher than that of 5%,20% respectively(P0.01).The effective intervention time was 72 h,and effective intervention concentration was 10%.After intervention of 72 h by 10% drug-containing,the OD values of chondrocyte in groups of serum containing Tougu Xiaotong capsule were significantly higher than that of blank group respectively(P0.01).The proliferative effect was the most obvious in experiment group 2(P0.05).Conclusion Serum containing Tougu Xiaotong capsule can promote the proliferation of chondrocyte,retain the activity of chondrocyte,and the proliferative effect is concerned with the drug concentration.In middle dose group of serum containing Tougu Xiaotong capsule,the proliferative effect is the best.
