Abstract
[Objective] To obtain optimum processing parameters for extraction of total flavonoids from Hippophae rhamnoides.[Method] H.rhamnoides from Qinhe Xinjiang was taken as study object,ultrasonic followed by microwave were used for the extraction of total flavonoids.First single-factor experiments were taken,such as ultrasonic time,ultrasonic temperature,ethanol concentration,microwave power,microwave time and so on.According to the results of single-factor experiments response surface methodology(RSM) was applied to optimized the extraction result and a maximum extraction yield of total flavonoids was achieved;Then compared with the results of ethanol refluxing extraction,microwave assisted extraction and ultrasonic assisted extraction.[Result] The optimum conditions shows: ultrasound conditions remain unchanged(ie,ultrasonic time 25 min,ultrasonic temperature 60 ℃) and 17.7-fold volume of 68.1% ethanol followed by ultrasonic treatment for 25 min and subsequent 80% microwave treatment for 3.33 min.Under these conditions,the content of sea buckthorn leaves flavonoids reached 42.75 mg/g.Compared with those the yield of total flavonoids was higher than 14.67%,24.04%,36.63%.[Conclusion] The study will provide a new idea for extraction of total flavonoids from H.Rhamnoides.
