Protective Effects of Tumor Necrosis Factor α Antisense Oligodeoxyrbonucleotide on Liver Ischemia Reperfusion in Rat

Abstract

Objective: To study the protective effects of tumor necrosis factor a (TNFa) antisense oligodeoxyrbonucleotide (ASODN)on liver ischemia reperfusion in rat. Methods: 160 male SD rats were randomly divided into four groups: normal control group, ischemia group, TNFa mismatching ODN group and TNFα ASODN group. The animals were killed at 1, 3, 12 h after 1h ischemia and followed reperfusion. Serum alanine aminotransferase (ALT), malodialdehyde (MDA) and liver histopathology were observed. Results: After ischemia reperfusion, the serum ALT and MDA were remarkable increased in ischemia group and TNFα mismathcing ODN group. The liver congestion was obviously. Treatment of TNFα ASODN could decrease serum ALT and MDA levels significantly and relieve hepatic congestion. Conclusion: TNFα ASODN could suppress the inflammatory reaction induced by hepatic ischemia reperfusion and has protective effects on rat hepatic ischemia reperfusion injury.
