Abstract
Objective:To investigate the effects of platelet-rich plasma(PRP) on the proliferation and osteogenic differentiation of rat muscle satellite cells(MSCs) in vitro,and explore the potential mechanism by which PRP affects MSCs.Methods:Blood samples drawn from heart of Sprague-Dawley rats were used to prepare PRP(Landesberg method). Platelet counts in PRP and whole blood were measured.The skeletal muscles of 3~5 d newborn SD rats were acquired to separate MSCs with enzyme digestion and differential centrifugalization technique.Myogenic markers desmin andα-sarcomeric actin in cytoplasm were detected by immunohistochemistry.The effects of different PRP concentrations(1.6%, 6.5%and 12.5%) on MSCs proliferation were assayed with MTT.Effect of PRP on the proliferation and differentiation of MSCs was measured by alkaline phosphatase(ALP) staining,Alizarin red S staining,osteocalcin and typeⅠcollagen immunohistochemistry staining.Results:6.25%PRP could significantly promote osteoblast proliferation.The MSCs in the PRP group was positive to ALP,osteocalcin and typeⅠcollagen.Calcium nodules could be seen after Alizarin red S staining.There was a significant increase in ALP activity of the PRP group compared to the control group(P0.01). Conclusion:PRP could obviously promote the proliferation and osteogenic differentiation of rat skeletal muscle satellite cells in vitro.
