Effect of Yishen Decoction on Renal Transforming Growth Factor β_1 Expression in Mice with Experimental IgA Nephropathy

Abstract

Objective To observe the effect of Yishen Decoction(YD) on renal transforming growth factor β1(TGF-β1)expression in mice with experimental IgA nephropathy.Methods BALB/C mice were used for the experimental animal,mice models of IgA nephropathy were induced by oral use of bovine serum albumin combined with injection of staphylococcal enterotoxin B into caudal vein.Renal periodic acid-schiff(PAS) stain,immunofluorescence method and 24-hour urinary protein quantitative examination were used to confirm the success of modeling.Then the mice models of IgA nephropathy were randomized into the model group,and high-and low-dose YD(0.05 and0.01g·kg-1·d-1 respectively)groups.A normal control group was also set up.Fluorescence quantitative polymerase chain reaction(FQ-PCR) was applied for the detection of renal TGF-β1 mRNA expression,and immunohistochemical assay was used for the examination of TGF-β1 content.Results Renal TGF-β1 mRNA expression and TGF-β1 content were increased in the model group(P0.05 compared with the normal control group),and were decreased in high-and low-dose YD groups(P0.05 compared with the model group).Conclusion The therapeutic mechanism of Yishen Decoction for IgA nephropathy is related with the inhibition of TGF-β1 secretion and mRNA expression.
