Abstract

AIM: To observe the effects of Porphyromonas gingivalis on expression of IL-6 and IL-1β in human umbilical vein endothelial cells(HUVECs).METHODS: Primary clutures of HUVECs were derived by enzyme digestion method.Passage 5 HUVECs were stimulated by P.gingivalis supernatant for 6,12,18 and 24 h.At specific time points,mRNA was extracted and subjected for RT-PCR to detect IL-6 and IL-1β mRNA expression.HUVEC culture supernatant was collected and subjected for ELISA to measure IL-6 and IL-1β protein levels.RESULTS: P.gingivalis supernatant stimulation significantly increased IL-1β mRNA expression at 12 and 18 h(P0.05),and it also significantly increased IL-6 mRNA at 12 h(P0.05).Furthermore,P.gingivalis stimulation resulted in significantly higher IL-1β secretion at 18 and 24 h(P0.05).And IL-6 protein level was significantly augmented at 12,18 and 24 h(P0.05).CONCLUSION: P.gingivalis can stimulate the expression of IL-6 and IL-1β in human umbilical vein endothelial cells.
