Effect of Alendronate on Proliferation of Human Osteolbasts

Abstract

Objective:Using modified digestive method to obtain and culture human osteoblast,and investigate the effect of alendronate on osteoblasts.Methods:Human ilium cancellous bone was harvested in surgery and digested by trypsin and collagenase of 30 mins to remove blood cells and fibroblast.Bone chips were digested again for l h,a large amount of pure osteoblasts were released and used for research.After 4 generation subculture,osteoblasts were seeded in 96-well plates at the density of 1×10 5/ml.24h later,10 7 M~10 3 M alendronate culture medium were changed.MMT method was used to assay proliferation of osteoblasts.Results:≥10 4 M alendronate inhibited proliferation of osteoblast;≤10 5 M alendronate has no effect on osteoblastic proliferation.Conclusion:The modified digestion method could obtain nearly pure human osteoblast.High concentrated alendronate inhibited osteoblastic proliferation.
