Screening of Lignin-degrading Fungi and Study of Lignin Degradation of Vineyard Pruning

Abstract

Lignin-degrading microorganisms with higher Lignin-degrading enzymes activites and higher Lignin degradation ability were isolated and selected from the decayed prunings collected in a vineyard.Twenty-four fungi strains which showed color zones on the culture medium of guaiacol plates were obtained by isolation and purification.Five strains with higher Lignin-degrading enzymes activity were screened by colorful reactions of PDA-Guaiacol plate and decolorizateion of PDA-Aniline bule plate.Liquid fermentation test and solid-state fermentation test of the five strains were taken.Results showed that A-51-1 was the most excellent strain with the higher lignin-degrading enzymes activities,for its laccase and Manganese-dependent peroxidase activities were up to 9.20 U·mL-1 and 21.6 U·mL-1 respectively;After treatment of vineyard prunings by A-51-1 for 30 d,the degradation rates of lignin reached 32.53%.
