Abstract
Objective To investigate the influence of IL-27 on serum IgE of asthmatic mice challenged by ovalbumin(OVA).Methods 24 female BALB/c mice were randomly divided into physiological saline group,asthmatic group and IL-27 group with each group of eight ones.The mice of IL-27 group were administered with 1 μg intranasal IL-27(dissolved in 50 μL PBS) after the establishment of asthmatic model with the application of OVA.The serum IL-4 and IFN-γ concentration of mice were detected with ELISA,the total serum IgE and OVA specific IgE concentration of mice were detected with fluorescence enzyme linked immunosorbent assay and the number of eosinophil granulocytes in bronchoalveolar lavage fluid(BALF) was counted.Results The serum IL-4 concentration [(45.6±10.1) μg/L] of mice in IL-27 group was significantly lower than that of asthmatic group [(79.3±14.8) μg/L](P0.05).The serum IFN-γ concentration [(63.8±11.5) μg/L]of mice in IL-27 group was obviously higher than that of the asthmatic group [(25.7±8.2) μg/L](P0.05).The total serum IgE [(2.77±0.51) KU/L] and OVA specific IgE concentration [(0.48±0.09) KU/L]of the mice in IL-27group were significantly lower than those [(5.88±0.86) KU/L,(1.89±0.15)KU/L] of the asthmatic group(both P0.05).The BALF eosinophil granulocytes counts [(2.21±0.33)×107/L]of the mice in IL-27 group was significantly lower than that of the asthmatic group [(12.82±2.17)×107/L](P0.01).Conclusion IL-27 reduces total serum IgE and OVA specific IgE concentration,the number of eosinophil granulocytes in BALF of asthmatic mice through promoting IFN-γ synthesis and inhibits IL-4 synthesis.
