Abstract

The clones of three kinds of cutting rose were constructed by tissue culture to explore the suitable culture medium for rapid propagation of cutting rose and provide a theoretical basis for mass propagation of cutting rose seedlings.The results showed that the preferable media for producing buds was 1/2 MS+6-BA 1.5 mg/L+NAA 0.5 mg for kalola(budding rate 81%),MS+6-BA 0.8 mg/L+NAA 0.1 mg/L for pink(budding rate 79%) and MS+6-BA 1.0 mg/L+NAA 0.1 mg/L for snow(budding rate 75%).The optimum multiplication medium for kalola,pink and snow were 1/2 MS+6-BA 1.0 mg/L+NAA 0.3 mg/L(multiplication coefficient 4.67),MS+6-BA 0.5 mg/L+NAA 0.1 mg/L(multiplication coefficient 3.15) and MS+6-BA 1.0 mg/L+NAA 0.05 mg/L(multiplication coefficient 3.33) respectively.1/2 MS+NAA 0.5 mg/L+IBA 0.1 mg/L was the optimum rooting medium for kalola;MS+NAA 0.1 mg/L+IBA 0.1 mg/L was the optimum rooting medium for pink;1/2 MS +NAA 0.5 mg/L was the optimum rooting medium for snow.Transplanting survival rate was all obviously high with vermiculite as media.
