Abstract
Objective To study effects of the lead exposure on activity of nitric oxide synthase(NOS)and concentration level of nitric oxide(NO)in rats spleen and their correlation with the blood lead.Methods Wistar rats were exposed to lead by drinking 0,18 4,and 184 mg/L lead acetate solution as the control group,and low and high dose group respectively.Activities of NOS and SOD,and concentration levels of NO and MDA were measured.Results Compared with the control group and low dose group,spleen NOS activity in high dose group lowered significantly at the time of 7 d (P0 05) ,but became significantly higher at 90 d (P0 05) .At 90 d ,spleen NOS activity in low dose group showed higher (P0 05) .As to spleen NO level,those in high and low dose groups were significantly higher than in the control group at 60 and 90 d (P0 05) .SOD activity in high dose group lowered at 14,30,60 and 90 d (P0 05) ,and the same results showed in low dose group at 30 and 90 d .MDA level in high dose group was remarkably increased at 14,60 and 90 d (P0 05) compared with the control group,but significantly lower than in low dose group at the time of 60 d (P0 05) .Parabola type,converse S type and converse parabola type dose effect curves existed for the relationship between the blood lead concentration and spleen MDA,NO level and SOD activity.Conclusions It indicated that activities of NOS and SOD,and the concentration levels of NO and MDA in rats spleen could be changed after lead exposure causing lipid per oxidization damage.
