Abstract
Objective To investigate the effect of rosiglitazone on severe acute pancreatitis(SAP)-associated lung injury in rats.Methods Fifty-four Wistar rats were randomly divided into three groups:control group,and SAP group,and SAP with rosiglitazone preconditioning group,18 rats in each group.SAP was induced by retrograde injection of 5% sodium taurocholate(0.1 ml/100 g)into the pancreatic duct.Control group was retrogradely injected sodium chloride(0.1 ml/100 g)into the pancreatic duct.Rosiglitazone group:the same as SAP,but rosiglitazone(6 mg/kg)was administered via femoral vein 30 min before operation.The expressions of NF-κB and MMP-9 in lungs were evaluated by immunohistochemical method.Serum amylase level,wet/dry weight ratio and histologic scoring of lung injury were determined.Results NF-κB and MMP-9 were hardly expressed in control group.The expressions of NF-κB and MMP-9 in SAP group were associated with time and peaked in SAP 12 h group.Application of rosiglitazone inhibited the activity of NF-κB and depressed the expression of MMP-9,reduced serum amylase level and wet/dry weight ratio of lung,and alleviated lung injury in 6,12 h group(P0.01).Conclusions Expressions of NF-κB and MMP-9 have close relation with lung injury in SAP.Application of rosiglitazone relieves the pathophysiological process.The potential mechanism is associated with depression of MMP-9 expression,and is most likely through the inhibition of NF-κB activity.
