Abstract
OBJECTIVE To analyze and investigate the influence of determination of the percentage of reticulocyte in mouse by Flow Cytometry and develop a rapid,accurate and stable flow cytometry assay for mouse reticulocyte percentage by optimazing key experiment parameter. METHODS Biological activity of rh EPO in vivo was determined according to reticulocyte method in European Pharmacopoeia 7. 0. RESULTS Individual differences of experiment animal,reaction time after exposure to different rh EPO dose and diffirences between instruments could effect the accuracy and pricision of assay result,but the error could reduced by optimizing parameters and taking some treatment measures. CONCLUSION A rapid,accurate and stable flow cytometry assay for mouse reticulocyte percentage can be accomplished after optimazing.
