Relationship of Visceral Fat Tissue and Insulin Sensitivity in Obesity Rats Induced by High-Fat Diet

Abstract

Objective:To observe the relationship of insulin sensitivity and the changes of fasting plasma TNF,α,FFAs,VISURAL FAT in obesity rats induced by high-fat diet. Methods: Normal male Wistar rats(9 weeks old) were divided into two groups taking either normal chow (NC,3.3Kcal/g,15~20% calories for fat),or high-fat diet (HF,5.1 Kcal/g,61% calories for fat).10 weeks later, body weight, visceral fat mass,fasting glucose,fasting insulin,TNF,αand FFAs were determined,and HOMA-IR was calculated by the method HOMA-IR=FPG×FINS/22.5. Results: Rats of HF showed significantly body weight and visceral fat mass gain (body weight increased by 11.4% in HF compared with that in NC,P0.05,visceral fat mass increased by 20.7% in HF compared with that in NC,P0.01) and developed the insulin resistance(HOMA-IR:HF: 8.88±4.25 vs NC:3.92±2.26,P0.05) after 10 weeks feeding. The concentration of fasting plasma FFAs and TNF,α were also increased significantly in HF group compared with NC group by 80.8% and 58.4%(P0.05).But there were no differences in fasting plasma glucose in the two groups(HF:7.89±1.46mmol/L vs NC:8.70±1.59mmol/L,P0.05).It demonstrated that VF was positively correlated with HOMA-IR(P0.05) in partial correlation analysis. Conclusion: In this study, obesity rat with insulin resistance has induced successfully with high-fat diet, and the state of insulin resistance of the rats has correlation with the increased mass of visceral fat and concentration of fasting plasma FFAs and TNF,α.
