Separation of human serum proteins by capillary electrophoresis

Abstract

This study details the qualitative results from a comparative evaluation of the separation of human serum proteins by cellulose acetate electrophoresis and free zone capillary electrophoresis.When the conditions for the separation of serum proteins were set at 100 mmol/L borate buffer at pH 98 in an untreated 50 μm(id)×21cm(16 cm to detector)capillary with detection wavelength of 200 nm.Five serum proteins were separated by the capillary electrophoresis in less than 5 minutes.The method had execellent reproducibility.Ten protein peaks were detected when a diluent containing 25 mmol/L sodium dodecyl sulfate (SDS),20 mmol/L borate buffer at pH 85 was used to dilute the serum samples in a 45 cm×50 μm(id)fusedsilica capillary(40 cm to detector)with a running voltage of 225 kVIn conclusion,the rapid analysis,execellent reproducibility of this automated capillary zone electrophoresis(CZE)method for the separation of serum proteins appears to be suitable for use in the routine laboratories in a clinical setting.
