Abstract
Microscope,AO-EB fluorescent staining, flow cytometry and DNA agarose gelelctrophoresis were used. SMMC-7721 cells incubated in serum-free RPMI 1640 medium for 48 h and then treated iwth 10#mu# mol/L C_2-ceramide for 24h. C_2-ceramide causes cell death in a dose and time-dependent manner. The apoptotic body and condensation of chromatin were detected with AO-EB fluorescent staining. The apoptotic peak was seen by the DNA content assay with flow cytometer. DNA agarose gel electrophorsis revealed DNA fragmentation and the appearance of the DNA ladder. C_2-ceramide can successfully induce apoptosis of hepatoma cells SMMC-7721.
