Abstract

Objective To observe the effect of C\-2\|ceramide on apoptosis of hepatoma cells SMMC\|7721. Methods Microscope, AO\|EB fluorescent staining, flow cytometry and agarose gel electrophoresis were used. Results SMMC\|7721 cells incubated in serum\|free RPMI 1640 medium for 48 h and then treated with 10 μmol/L C\-2\|ceramide for 24 h. C\-2\|ceramide causes cell death in a dose and time\|dependent manner. The apoptotic body and condensation of chromatin were detected with AO\|EB fluorescent staining. The apoptotic peak was seen by the content assay with flow cytometer. agarose gel electrophoresis revealed fragmentation and the appearance of the DNA ladder. Conclusion C\-2\|ceramide can successfully induce apoptosis of hepatoma cells SMMC\|7721.
