Abstract
In order to improve the enzyme activity of cold temperature amylase,the technological conditions were used to optimize Bacillus subtilis to probuce cold temperature amylase by response surface.Based on single factor experiment,the 3 factors affacting enzyme activity were as follows:the best appropriate temperature,the best appropriate pH value and metal ions.The enzyme activity was used as response value to respond to surface optimization and to test the optimized plan.The experiment result showed that the optimized parameters of enzyme activity of cold temperature amylase were:the most appropriate response temperature was 30 ℃,the most appropriate pH value was 6,the metal ions with the strongest activation for enzyme were Ca2+,whose concentration was 0.01 mol/L,under above conditions,the activity of cold temperature amylase was 32.67 U/mL,which was the good match for the model.
