Effects of IL-17 on proliferation and invasion of B16 F10 melanoma cells

Abstract

Objective: To investigate the influence of interlukin-17(IL-17) on the proliferation and invasion of mouse B16F10 melanoma cells.Methods: Mouse B16 melanoma cells were cultured in vitro.Cells in research group were treatead with a certain-concentration IL-17 and those in control group treated without IL-17.The proliferation of B16 cells was detected by MTT.And the invasieness of B16 cells was determined by t ranswell.Then the expression activities of MMP-2 and MMP-9 of cells were mesured by Gelatin Zymography.Results: The growth rate of B16 cells treated with IL-17 was not changed(P0.05) compared with the control group.Matrigel invasion assay revealed that addition of IL-17 to B16 cells stimulated marked invasion(P0.01).And Gelatin Zymography analyzed that the expression of MMP-2 and MMP-9 of cells was significantly enhanced in the research group(P0.01).Conclusion: IL-17 has no effect on proliferation of B16F10 cells but can promote the invasion which may due to the increased MMP-2 and MMP-9 activities.
