Influences of puerarin on content of ICAM-1,VCAM-1,MCP-1,IL-6 and NO in human umbilical vein endothelial cells

Abstract

Objective To observe the influences of puerarin on the content of intercellular adhesion molecule-1(ICAM-1),vascular cell adhension molecule-1(VCAM-1),monocyte chemoattractant protein-1(MCP-1),interleukin-6(IL-6) and nitric oxide(NO).Methods Human umbilical vein endothelial cells(HUVE) were collected and divided into the normal group,model group,puerarin group and Rapamycin group.The model group was stimulated by thrombin in a final concentration(5 u/L).The growth of HUVE was detected by using MTT and flow cytometry.The expressions of IL-6,MCP-1,ICAM-1 and VCAM-1 were examined by applying ELISA and NO expression was determined by using the method of nitrate reductase.Results The outcomes of MTT detection showed that when the concentration of puerarin was in 1.0×10-2 mol/L or 1.0×10-4,it inhibited significantly the growth of HUVE stimulated by thrombin(P0.01).The outcomes of flow cytometry showed that,compared with the model group,the phases of S and G2 induced by thrombin were inhibited(P0.05),the expressions of IL-6,MCP-1,ICAM-1 and VCAM-1 decreased significantly(P0.05),NO content increased significantly(P0.01),and expression of ICAM-1 was not changed(P0.05) in the puerarin group.Conclusion Puerarin can inhibit the division and proliferation of vascular endothelial cells induced by thrombin,which is manifested as inhibition of phases of S and G2.The mechanism may be related to the increase of NO content,and inhibition of the release of VCAM-1,MCP-1 and IL-6 by vascular endothelial cells.
