Effect of intravenous transplantation of bone marrow mesenchymal stem cells on rat acute myocardial infarction

Abstract

Objective To investigate the effect of intravenous transplantation of bone marrow mesenchymal stem cells(MSCs) for one time and many times on acute myocardial infarction(AMI) in rats. Methods Sixty rats were randomly divided into group A(iv transplantation for one time), group B(iv transplantation for many times),group C(control of iv transplantation for one time) and group D(control of iv transplantation for many times) of 15 rats each. 5×10~6 labeled MSCs were injected to AMI rats on day 1(group A) and days 1,4,7,10,13(group B) after MI,and culture medium of the same volume was injected to groups C and D. Four weeks after MI,the cardiac function was evaluated and immunohistochemical analysis was performed. Results Many DAPI positive cells could be found in infarcted myocardium of groups A and B. Compared with groups C and D,the left ventricular function was obviously improved and capillary density was obviously increased while infarct size was significantly reduced in groups A and B (P0.05). There was significant difference between groups A and B (P0.05). Conclusion The results demonstrated that iv transplantation of bone marrow mesenchymal stem cells for many times has better effect on rats with AMI compared with transplantation for one time.
