Inhibiting Kupffer's cells attenuates the reperfusion injury of hepatocellular mitochondria

Abstract

Objective:To research the effect on reperfusion injury of hepatocellular mitochondria in rats by inhibition of kupffer cells.Methods:The rats in experimental group were pretreated with Gadolinium(7mg/kg) by introveinal injection for 2 days,and the rats in the control group were treated with saline solution.Two,six and eighteen hours of reperfusion of the liver arter 2 hr of ischemia were performed by operation on the third day.The concentrations of alanine aminotransferase(ALT),of malondialdehyde(MDA)in hepatic tissue and hepatocellular mitochondria were determined.The changes of morphology of hepatic tissue in optical and ultrastructure of hepatocyte and hepatocellular mitochongdria were observed.Results:The contents of MDA of hepatocellular mitochondria in 2 and 6 hours of reperfusion were (1.81±0.24)nmol/g and (1.91±0.56)nmol/g respectively in experimental group,and the contents of MDA of hepatic tissue in 6 hours of reperfusion in experimenta group was (3.50±2.06) nmol/g,and the results in contrast group were (4.89±2.91) nmol/g(P0 05),(2.93±0.51) nmol/g(P0 05) and (7.65±2.46) nmol/g (P0 05) respectively.With the prolongation of the time of reperfusion,karyopyknosis,condensed chromation near nuclear membrane and decreased subcellular structures could be observed,and hepatocyte be demolished when the injury was severe in contrast group,while general hepatocellular appearance was still observed even though in 18 hours of reperfusion in experimental group.And the extent of injury in mitochondria was evidently aba ted.Conclusion:Inhibiting Kupffer's cells can attenuate the reperfusion injury of hepatocellular mitochondria.
