Embryogenic callus induction and somatic embryogenesis of papaya

Abstract

This test regards immature embryos,hypocotyl,leaves and cotyledons of papaya(Carica papaya L.) as explants,studying and exploring the effect of hormones,attachment,developing methods and conditions on the papaya embryogenic calli and somatic embryos induction.The experimental results showed that immature embryo was suit to somatic embryogenesis.The best medium for callus induction was modified MS+10 mg·L-1 2,4-D+5 mg·L-1 NAA+2 mg·L-1 KT+0.5 mg·L-1 BA,the most suitable medium for callus proliferation was modified MS+10 mg·L-1 2,4-D+2.5 mg·L-1 NAA+1 mg·L-1 KT+0.5 mg·L-1 BA,and the best medium of somatic embryogenesis was modified MS+10 mg·L-1 2,4-D+2 mg·L-1 KT.Embryogenic calli induction from papaya cotyledon was difficult,calli induction from leaves or hypocotyl was easy,but somatic embryogenesis was very difficult.
