Abstract
Objective:To investigate the effects of Topiramate on rat cognitive function and the content of serotonin(5-HT) and acetylcholine esterase(AChE) activity in the rat hippocampus.Methods:The Wistar rats aged 28 days were randomly divided into 3 groups.TPM were consecutively used for 28 days.Every group was subjected to the Y-maze testing to examine the learning and memory.The AChE activity in the rat hippocampus was measured by chemicolorimetric method,while the 5-HT expression in the rat hippocampus was done by immunohistochemical method.Result:Compared TPM-control A and B groups with blank group,and compared TPM-control B group with TPM-control A group,the discrimination and 24h memory retention of the rats in Y-maze testing were obviously lowered(P0.01),the number of 5-HT positive neuron in immunoreactivity was significantly reduced(P0.01),and the AChE activity in the rat hippocampus was much higher(P0.01,P0.05).Conclusion:TPM was harmful to cognitive function,and the degree of cognitive functional impairment may be related to the dose of the drug,and the cognitive functional impairment caused by TPM may be correlated with the 5-HT decrease and the increase in AChE activity in rat hippocampus.
