Abstract
Objective:To investigate the association of S100A4 gene expression with differentiation,metastasis and prognosis of colorectal carcinoma.Methods:The SP immunohistochemical method was used to detect the expression of S100A4 in 87 cases of colorectal carcinoma and 87 cases of adjacent colorectal tissues.The relationship between the expression of S100A4 and the clinicopathologic factors and post-operative five-year survival rate was analyzed.The expression of S100A4 in three colorectal carcinoma cell lines Caco-2,HT29,HCT116 with different differentiation degrees was detected by using the SP immunocytochemical method;the mRNA expressions of S100A4 was measured in the colorectal carcinoma cell lines by using RT-PCR.Results:There was no S100A4 gene expression in glandular epithelium of adjacent colorectal tissues.The positive expression rate of S100A4 was 64.4%(56/87) in colorectal carcinoma;there was significant difference in S100A4 gene expression between colorectal carcinoma and adjacent colorectal tissues(P0.01).The expression of S100A4 positively correlated with the clinical stages,lymph node metastasis and five-year survival rate(P0.05),but not with other clinicopathologic factors(P0.05).S100A4 protein was expressed in the three colorectal carcinoma cell lines.The expression of S100A4 mRNA was the highest in HCT116 cells,moderate in HT29 cells,and the lowest in Caco-2 cells.There were significant differences in the expression level of S100A4 mRNA of the three cell lines(P0.01).Conclusion:S100A4 gene was closely related with initiation and progression,differentiation,invasion,metastasis and prognosis of colorectal cancer.S100A4 might be an important predictor of the clinicopathologic features and prognosis of colorectal carcinoma;and might be expected to become a new biomarker and potential target for colorectal carcinoma treatment.
