Abstract
Objective To explore the role of paraoxonase 1(PON1) gene Q192R polymorphism and activity in occurrence of cerebral infarction.Methods 104 patients with cerebral infarction were analyzed and divided into two groups(60 years and ≥60 years) of 63 patients and 41 patients respectively.104 subjects matched simultaneously in age and sex served as controls.The PON1 Q192R polymorphism was determined by polymerase chain reaction amplification and restriction digestion.Enzyme activity was determined by spectrophotometric assay.Results The genotype distribution of PON1 Q192R polymorphism was not different between the old cerebral infarction patients and the controls.The prevalence of the PON1 192RR genotype was significantly increased and PON1 192QQ genotype was significantly decreased in the young and middle-aged cerebral infarction patients compared with controls(χ2=11.730,P=0.003).PON1 activity of both groups of patients was lower than that of controls(P0.001).PON1 activity was negatively correlated with the frequency of the R allele in both old people and young or middle-aged people.High PON1 activity could decrease the risk of cerebral infarction in both old people(OR=0.823,95%CI:0.759-0.891,P0.001) and young or middle-aged people(OR=0.835,95%CI:0.788-0.885,P0.001).Conclusions PON1 192Q/R genotype can affect PON1 activity.High PON1 activity can prevent the development of cerebral infarction,but PON1 Q192R polymorphism has no effect on the development of cerebral infarction.
