Abstract
Objective:To study the effect of ginseng extract on Caspase-3in rabbit ear so as to investigate the prospect of hypertrophic scar treatment.Methods:15hypertrophic scar models were established,and 2to 3wounds in the ventral and middle ear of each rabbit were done randomly.Then they were divided into the blank control group(Group A),the saline injection group(Group B), and the ginsenoside Rb1groups(Group C,D and E).Group C,D,E were injected with 0.2mg/ml,0.4mg/ml and 1mg/ml ginsenoside Rb1.The wound healing and scar hyperplasia after operations were generally observed.Epithelization was conducted the corresponding treatment by group after about 26days by being administered once every three days,with a total of 3times.After having been administered,scar tissue specimens of the rabbits were taken on the 2nd,4th and 6th week respectively,4rabbits each time to have HE staining and Masson staining.Then HI and collagen fiber density were measured and calculated,and the expression of Caspase 3was detected by immunohistochemical measurement.Results:With the increase of drug concentrations of HI,collagen density decreased,but there was no difference between Group C and Group D(P0.05),and three were differences between Group E and Group C and D(P0.05).Histochemical results were also found in Caspase 3that Caspase-3expression increased with the intervention of high concentration of drugs.Conclusion:Rb1components of ginseng extract can reduce the synthesis of collagen in the scar tissue and induce the cell apoptosis,thus effectively inhibit hypertrophic scar.
