Abstract
Objective To evaluate the alteration of heat shock protein 70 (HSP 70) of lung during severe acute pancreatitis (SAP) and the therapeutic effects of ketamine in rats. Methods A total of 48 SD male rats were randomly divided into Group A (sham-operative group), Group B (SAP group) and Group C (SAP treated with ketamine), 16 animals in each group. The model of rat SAP induced by retrograde injection of 5% sodium taurocholate into the pancreatic duct of rats. And the treatment groups received ketmine with doses of 50 mg/(kg·h) injected continually immediately after the preparation of SAP were made. All rats were killed at 12 hours after operation. Bronchoalveolar lavage fluid (BALF) was collected for determination of pro-tein and PMN count. The lungs were removed for evaluation of HSP 70 and determination of wetPdry lung weight ratio. Results In SAP with ketamin therapy group, the levels of HSP 70 were significantly higher than those of SAP group (P 0.05), and the wetPdry lung weight ratio and the protein and PMN count in BALF were significantly lower than SAP group. ConclusionHSP 70 are increased remarkably in SAP. Ketamine has protective effect against severe acute pancreatitis associated lung injury in rats by inducing the peak level of HSP 70.
