A Study on the Effect of Antisense Oligonucleotide Targeting Survivin to the Proliferation and Apoptosis of A549 Cells

Abstract

Objective To investigate the effect of survivin antisense oligonucleotide to the proliferation and apoptosis of A549 cells. Methods Designed a human survivin antisense oligonucleotides and transfected it into A549 cells.The expression of survivin mRNA and the level of protein were detected by RT-PCR and Western blot methods respectively.The MTT assay was used to detect the cell proliferation and the apeptotic rates were measured by TUNEL assay.Results The quantity of survivin mRNA and the level of protein were decreased by this antisense oligonucleotide.The antisense oligonucleotides had a inhibitory rate of proliferation which was much higher than the rate of the control.The apoptotic rate obtained by transfecting cells with antisense oligonucleotide was higher than the rate of control which all exposed to the low concentration of homoharringtonine.Conclusion The experiment results indicate that antisense oligonneleotide can deserve as an approach to subservient tumor therapy in future.
