Abstract
Ning-xia lycium barbarum was used as raw material for the purpose of polysaccharide extraction with water.Glucose was the control sample,the content of lycium barbarum polysaccharide was determined by phenol-sulphoacid method.Then the orthogonal layout L9(34)was used,the extraction technology of lycium barbarum polysaccharide was optimized by orthogonal experimentation of 3 factors and 3 levels,factors of temperature,extraction times and solid-liquid ratio were evaluated for their effects on extraction process.According to the orthogonal experimental design,the optimum extracting conditions of polysaccharide from lycium barbarum was showed as following: extracting temperature was 50℃,solid-liquid ratio was 1∶30,extracting time was 2h.The extraction rate of lycium barbarum polysaccharide reached 4.88% with the conditions of optimum extraction.The average recovery was 97.97%,relative standard deviation RSD was 0.74%(n = 5).
