Abstract

Objective To establish a method of constructing a bilayered living skin equivalent in vitro.Methods Using serially passaged keratinocytes and dermal fibroblasts as seed cells type I collagen as matrix,living skin equivalent has been generated through air-liquid interface culture.Sections of skin equivalent were stained with HE and examined under microscopy.Results Morphology of skin equivalent was similar to that of normal skin.The skin equivalent has density dermis and good differentiated epidermis after cultured in air-liquid interface.Conclusion The artificial skin made by us is a satisfactory skin substitute with normal dermis and epidermis.The method employed here can be used to fabricate living skin equivalent.[
