Abstract

Objective To investigate the renal tubule interstitial fibrosis in obstructive nephropathy rats.Methods In adult SD rats,left ureter was ligated to create the model of obstructive nephropathy (UUO).Rats with UUO were sacrificed 3,7,10,and 14 days after obstruction.By using immunohistochemistry,α smooth muscle actin (α-SMA) deposit and the counts of Monocyte/macrophage infiltrating into kidney were analyzed.The expression of transforming growth factor β1 (TGF-β1) mRNA,TIMP-1 mRNA and Col-IV mRNA was detected by using reverse transcription polymerase chain reaction (RT-PCR).Results The deposit of α-SMA,the counts of monocyte/macrophage infiltrating into kidney,the expression of Col-IV mRNA and TIMP-1 mRNA were increased after renal obstruction,suggesting the progressive renal tubule interstitial fibrosis.TGF-β1 was also increased (P0.05).Conclusion TGF-β1 promoted the renal tubule interstitial fibrosis in rats with UUO.
