Investigation of Simiaoyong'an decoction on IL-6,TNF-α and VEGF in human umbilical vein endothelial cells with anoxia

Abstract

Objective To investigate the effect of Simiaoyong'an decoction on IL-6,TNF- αand VEGF in human umbilical vein endothelial cells( HUVEC) with anoxia. Methods Human umbilical vein endothelial( HUVEC) cell lines were subcultured. The cells were divided into three experimental groups,the normal group,model group,Simiaoyong'an decoction group. The normal HUVEC in the normal group were cultured in normal culture solution. HUVEC in model group received anaerobic treatment and then were cultured by normal culture solution. HUVEC in Simiaoyong'an decoction group received anaerobic treatment and then were cultured by 10% Simiaoyong'an decoction culture solution. The levels of IL- 6,TNF- a and VEGF were measured by ELISA and immunohistochemistry in each group. Results There were significant differences between model group,Simiaoyong'an decoction group and the normal group on the levels of IL- 6,TNF- a and VEGF( P 0. 01). The levels of IL- 6 and TNF- αwas increased,and VEGF was decreased. But the increase of IL- 6 and TNF- αand the decrease of VEGF in Simiaoyong'an decoction group was less than that in model group( P 0. 01). Simiaoyong'an decoction can improve the levels of IL- 6,TNF- a and VEGF in HUVEC with anoxia. Conclusion Simiaoyong'an decoction can effectively inhibit the secretion of inflammatory factors in human umbilical vein endothelial cells with anoxia,promote the proliferation of endothelial cells,have anti- inflammatory and angiogenic effects on vascular endothelial cell injury has a protective effect.
