Abstract
A reversed-phase high performance liquid chromatography was developed for the determination of nucleotides (uridine, guanosine, adenosine and adenine)in cordyceps. The nucleotides were well separated and determined on an Alltima C18 column (250mm×4.6mm, 5μm)by using a mixture of acetonitrile-water (V/V=4:96) at 260nm with the flow rate of 1.0ml/min. The results showed that the developed method was sensitive, accurate, reproducible and high recovery(98.7%~101.5%) and suitable for analysis of nucleotides in cordyceps. The contents of uridine, guanosine, adenosine and adenine were ~10.52 mg/g,~8.56 mg/g,~0.708 mg/g and ~1.79 mg/g, respectively, with the average RSD ranged between 0.51% and 1.30% (n=5).
