Abstract

Objective To investigate the effect of salidroside on the proliferation and apoptosis of human melanoma cell line A375. Methods The A375 cells were cultured with different concentrations of salidroside( 0,0. 01,0. 1,1,10,and 100 μg /mL). The inhibition of cellular proliferation was determined by MTT. The apoptosis was assessed by the Annexin-V-FIFT /PI affinity assay. The mRNA levels of CDk4,CDk2,cyclinE and cyclinD1 were detected by RT-PCR. Results Compared with the negative control group,the proliferation inhibition rate of A375 cells of the salidroside group was significantly higher( P 0. 05). The cells of the salidroside group were mainly at the G0/G1phase,and the cells increased in the S phase and decreased in the G2/M phase( P 0. 05). The mRNA expression of CDk2,CDk4,cyclinD1and cyclinE in the salidroside group was inhibited,which indicated that salidroside limited the proliferation of A375 cells by regulating the cell cycle,and the difference was statistically significant( P 0. 05). Conclusion Salidroside has the effect of inhibiting the proliferation and enhancing the apoptosis of human melanoma cell line A375.
