Abstract

Foreign gene phy A was transfered to embryonic callus of maize inbred lines GS01 with Agrobacterium EHA105-mediated. Herbicides(alachlor P) were used to screen resistant callus and regenerate plantlets. The results showed that bacteria concentration and infection time affected callus conversion rate significantly. Types of hormone and proportion affected obviously regeneration plantlet of resistant callus. Bacteria concentration was 0.6 at OD600 nm. Infection time was 20 min. Resistant callus rate was high with 68.7%. It will provide a good reference for the regeneration and transformation of maize mediated by Agrobacterium.
