Optimization of ethanol extraction process of ‘Ershen pills 'drug pair by orthogonal test based on UPLC fingerprints

Abstract

OBJECTIVE To develop an UPLC fingerprint combined with orthogonal experiment design to optimize the best alcohol extraction process of‘Ershen pills' drug pair. METHODS L9(34) orthogonal experiment was used with ethanol concentration,dosage of ethanol,extraction time and extraction times as four factors,UPLC fingerprint characteristic peak area,contents of psoralen,isopsoralen,dehydrodiisoeugenol reference,and dry extract yield as comprehensive evaluation indexes. RESULTS Optimization of the best alcohol extraction process of‘Ershen pills' drug pair: 60% ethanol concentration,8 volumes of ethanol,extraction for 2times,1 h for each time. CONCLUSION Optimized extraction process is stable,reasonable and highly efficient,and has certain significance for regulation and quality control of alcohol extraction process of Ershen pills drug pair.
