Abstract

Objective To study the function of the new gene p15rs on the proliferation of HeLa cells,which is related to CKIp15 INK4B. Methods The coding region of p15rs was cloned and transfected into HeLa cells.The growth curve,cell cycle and colony forming of the cell model were tested. Results The cell model HPS21 overexpressing p15rs was constructed.The overexpression of p15rs inhibited the growth rate of HeLa cell obviously.The double time of proliferation in HPS21 was delayed.Compared with control HPC4,HPS21 cells in G1 phase increased but which in S phase decreased.Furthermore,the capacity of growth in soft agar of HPS21 was strongly decreased.Conclusion The new gene p15rs may suppress the proliferation of tumor cell and negatively regulate the cell cycle.
